Must say that i'm not impressed with the controls (on the PC version) - tried an XBox 360 wireless pad but that's just a pain with this game. Tried keyboard + mouse but just can't get the mouse sensitivity right: it's either too twitchy or too sluggish.
I managed to find a comfortable setting just a little bit higher than the default. Have you tried it with mouse acceleration off and on to see if you prefer that? (Personally I like it off, but YMMV.)
I also found that the comfort of the mouse controls was directly proportionate to my framerate, with everything totally maxed out I was regularly down at around 30FPS which made the mouse feel a bit twitchy, but dropping the shadows down two notches to HIGH literally doubled that to a steady 60FPS+, which made the controls a lot smoother and far more comfortable. (Everything else I could leave at the top settings.)
The graphics option screen is cleverly designed to be representative of about as demanding as the game engine gets, and all changes are made in real time, so you can immediately see what impact your changes are having on your framerate. If you can get 60FPS (or thereabouts) on the graphics options screen, that will relate to a pretty steady 60FPS in the game - and should help with the mouse controls. My experience was a lot better once I knocked the shadow options down from MEGA-SUPER-CRAZY-ULTRA to plain old HIGH (and if I'm entirely honest, I really had to look hard to see much of a difference).
FRAPS or MSI Afterburner will give you an OSD of your framerate.
